Once the double-stranded DNA is separated into single strands, the sequence of bases is exposed. The temperature is lowered to allow new bonds to form, and free floating primers in the solution begin to bind to complementary sequences on the open DNA strands. If your primers have been well designed, they will bind almost exclusively to the ends of the DNA fragment you hope to copy.
